turning the key the wrong way, would be my first guess.
Why do you need quick help though?

theres only a few options...
one: your turning the knob/lev er/deadbolt the wrong way to unlock
two: your lock is broken/damaged
three: you broke/damaged your lock...

Doors become tight when the foundations of the building settle, sometimes the weight of the door is resting on the bolt, lift the door by the handle and then try to turn it

I was thinking I needed to modify my key. Turns out I was practicing on it one way and did not realize different doors unlock different directions. (So obvious now) Hope this helps some other helpless fool like myself. Thanks!
Needed help quick because I lack patience and it was driving me nuts!

Almost right with the euro answer but not quite,
Its likely a euro cylinder but as youve removed the tip off your key it no longer pushes the cam in to oporate the mechs,
You really should not be doing it on your door although i suppose it gets work in

Certain Makes and Models do Matter as for a kwikset key in knob lock it unlocks a certain way even if it is upside down. and for lock picking some times it is easier to pick a lock in the wrong direction then to spin the plug back to the correct way.
