Yes, it will. I don't know the correct terminology, but some locks require a full revolution to unlock, meaning that you have to either pick the lock multiple times, though i've heard that a plug spinner turns the lock fast enough not to require that.
Hope this helps.

If it spins 180 and sticks: use the back of a half diamond to lift the tops pins out of the bottom of the keyway.

I think TOWCH is right. Normally when a key opens a lock, then lock will turn easily past that 180 degree point because the bottom of the key keeps those top pins from entering the keyway. When the key isn't present, the top pins are able to fall down the keyway. This isn't too much of a problem on regular cylinders, but it can be very bad if it is masterkeyed.
As TOWCH said, whenever you need to turn past that 180 degree point, always put the flat side of a half diamond pick in the bottom of the keyway to stop these top pins from falling down.

It doesn't have to be a diamond pick really. If I'm picking with a hook pick and the top pins fall into the bottom of the plug, I just apply medium tension and just run the tip of the pick from the back of the lock to the very front with the working tip of the pick to push the drivers back up to clear them.
If a lock is a puzzle, then its key is the complete picture

If its stopping at 180 degrees then its certainly what TOWCH said, and you only have to stroke a diamond alont the top of the (bottom actualy) keyway and the plug will continue turning, but if its not the top pins dropping into the lower open area of the keyway, it could be that you are picking a lock that is installed on a door, and there are issues involving the tailpiece inside the lock which could come from a bad installation or from being cheap, old and worn.
