Sounds like the bitting on the key is off by one cut somewhere. You *can* possibly turn it, but the pin the the plug is just barely clearing the cylinder housing and is compressed while you turn, giving a grinding feeling. That's my guess anyway. Does the key look normal or does it look cut by hand? I'm assuming this is the master. Does the submaster work at all?
One possible solution (if this is the problem) would be to go through the steps of impressioning the lock to find the spot on the key that's too high and file it down just a bit. This will change the key, so I recommend getting the key copied and trying this on the copy.
